Fenugreek Extract (4-HIL) Herbal Extraction Plants — Mechotech

Fenugreek Extract (4-HIL)

Purity Levels

40% Iso Lucin 4-HIL by HPLC

Mechotech designs and manufactures turnkey Fenugreek (Trigonella foenum-graecum) seed extraction plants standardised to 40% Iso Lucin 4-HIL by HPLC — a non-protein amino acid (4-hydroxyisoleucine) marker quantified by reverse-phase HPLC assay. This is a distinct grade from Mechotech's other Fenugreek Extract line, which is standardised to 40% Lucin Saponins by Gravimetry: that plant targets the steroidal saponin/diosgenin fraction and releases on a gravimetric assay, while this plant is tuned to concentrate and preserve the polar, heat-sensitive 4-HIL amino acid fraction and releases strictly on HPLC assay against a 4-HIL reference standard. We supply the complete machinery — seed cleaning, milling, aqueous/hydro-alcoholic extraction tuned for polar amino-acid recovery, filtration, low-temperature concentration, solvent recovery, standardisation and drying — for the pharmaceutical, nutraceutical, and food ingredient industries.

Mechotech's HPLC-grade Fenugreek plant is engineered around the distinct extraction behaviour of 4-hydroxyisoleucine: unlike the moderately non-polar steroidal saponins recovered by the gravimetric-grade line, 4-HIL is a small, highly water-soluble amino acid that partitions preferentially into the aqueous phase, is prone to loss in high-temperature evaporation, and requires closer control of extraction polarity (solvent-to-water ratio) and holding time to prevent degradation and to keep co-extracted sugars and pigments from suppressing the HPLC assay response. Extraction, concentration and drying parameters on this line are set specifically to maximise 4-HIL recovery and stability rather than saponin yield, and every batch is released only after HPLC assay against a 4-HIL reference standard — not the gravimetric method used for Mechotech's other Fenugreek Extract grade. Built in SS 316L with integrated solvent recovery, PLC-based automation and full batch documentation, each plant is delivered as a commissioned turnkey system to GMP norms.

Manufacturing Process

1

Raw Material Reception & Cleaning

Fenugreek seed is authenticated, cleaned of stones and foreign matter, tested for moisture, and graded before processing.

2

Grinding

Cleaned seed is milled to a controlled, uniform particle size to maximise contact area for the polar solvent system used in the following step.

3

Low-Temperature Aqueous/Hydro-Alcoholic Extraction

Milled seed is extracted in closed jacketed SS 316L extractors using a solvent-to-water ratio tuned toward the polar end of the range, under controlled low temperature and multiple cycles, to favour 4-hydroxyisoleucine recovery over the steroidal saponin fraction.

4

Filtration

The crude extract is clarified through pressure leaf or plate-and-frame filter presses followed by cartridge polish filters to remove suspended seed fibre and mucilage ahead of concentration.

5

Low-Temperature Vacuum Concentration & Solvent Recovery

The filtered extract is concentrated in a falling-film / vacuum evaporator held at a lower temperature band than the saponin-grade process to limit thermal degradation of the amino-acid fraction, with solvent recovered and recycled.

6

Colour & Sugar Reduction (Optional)

Where required, activated-carbon or resin polishing reduces co-extracted pigments and sugars that would otherwise interfere with HPLC baseline resolution of the 4-HIL peak.

7

Standardisation

The concentrate is assayed by HPLC against a 4-hydroxyisoleucine reference standard and adjusted to the declared 40% Iso Lucin 4-HIL by HPLC specification before drying.

8

Drying & Quality Control

The standardised extract is spray- or vacuum-dried under a low-temperature profile to a free-flowing powder, then released after HPLC assay, residual-solvent, heavy-metal, microbial, and moisture testing with full certificate-of-analysis documentation.

Frequently Asked Questions

Does Mechotech supply the complete machinery for this Fenugreek Extract grade?
Yes. Mechotech is a fenugreek extraction plant manufacturer and equipment supplier, engineering and fabricating the complete turnkey plant for the 40% Iso Lucin 4-HIL by HPLC grade at our Hyderabad facility, installing and commissioning it as a single integrated system.
How is this Fenugreek Extract plant different from Mechotech's other Fenugreek Extract page?
This is a different specification and a different process. This plant is standardised to 40% Iso Lucin 4-HIL by HPLC — the 4-hydroxyisoleucine amino-acid fraction, assayed by HPLC — and runs a lower-temperature, more polar extraction and concentration profile to protect that fraction. Mechotech's other Fenugreek Extract plant targets 40% Lucin Saponins by Gravimetry, the steroidal saponin fraction, released on a gravimetric assay instead. The two use different solvent polarity, temperature control, and release testing, so confirm which marker compound and assay method your buyer specification requires before selecting between them.
Why does the extraction temperature matter for a 4-HIL specification?
4-Hydroxyisoleucine is a small, water-soluble, heat-sensitive amino acid. Running extraction and vacuum concentration at the same temperature used for the steroidal-saponin grade risks degrading the 4-HIL fraction and lowering HPLC assay recovery, so this plant's thermal profile is deliberately set lower and its solvent system tuned more polar to favour 4-HIL over saponins.
What testing confirms the 40% Iso Lucin 4-HIL by HPLC specification?
Each batch is assayed by reverse-phase HPLC against a 4-hydroxyisoleucine reference standard, with optional activated-carbon or resin polishing beforehand to reduce pigment and sugar interference with peak resolution. Batches are released with a full certificate of analysis covering assay, residual solvent, heavy metals, microbial limits, and moisture.
What production capacities are available for this Fenugreek Extract plant?
Mechotech builds this Fenugreek Extract plant from pilot systems for product development to commercial-scale plants. We recommend a pilot plant to validate 4-HIL recovery and assay consistency before scaling.
Which industries use the HPLC-grade Fenugreek Extract plant?
Nutraceutical manufacturers making metabolic-health and blood-sugar-support supplements, sports-nutrition brands, and pharmaceutical buyers requiring HPLC-verified certificates of analysis are the primary users, alongside export markets across India, USA, UAE, and Nigeria.
Can Mechotech build both Fenugreek Extract grades on adjacent lines?
Yes — because the two grades draw on the same fenugreek seed but diverge at the extraction and concentration stage (polarity and temperature), Mechotech can engineer both the saponin-gravimetric line and this 4-HIL-HPLC line within the same facility, sharing common utilities while keeping extraction, concentration and drying equipment dedicated to each specification to avoid cross-contamination of assay results.
What does this Fenugreek Extract plant cost, and how is pricing structured?
Project cost depends on batch capacity, automation level, and whether optional colour/sugar polishing is included. Mechotech quotes a fixed turnkey price after reviewing your target capacity, HPLC specification, and site utilities; we recommend starting with a pilot plant to validate 4-HIL recovery and cost-per-kg before committing to commercial scale.
